In 1976, 11-year-old Gru tries to become a supervillain with the help of his minions, minions. His dream comes true when his application to join the ranks of the villainous organization "The Evil Six", headed by Wild Donna, is approved and he is invited to an interview. The "Six" are looking for a new member to replace their former leader, Grandfather Brass Knuckles, whom they fired and, as they believe, killed after stealing the Zodiac Stone, an ancient amulet associated with the Chinese zodiac. A certain Dr. Nefario helps Gru get to the secret base of the "six". The villains do not accept Gru into their ranks because of his age, and he, to their indignation, steals the amulet and escapes with the minions Kevin, Stuart and Bob, giving the amulet to another minion, Otto, for safekeeping.

Once in his lair, Gru finds out that Otto exchanged the amulet for a pet stone, as a result of which he becomes enraged, dismisses the minions and goes alone in search of the amulet. But as soon as Gru leaves the threshold of his house, Brass Knuckles, who turns out to be alive, kidnaps him, takes him to San Francisco and demands that the minions give him the amulet within forty-eight hours, otherwise Gru will be killed.

After an unsuccessful attempt to find the amulet, Kevin, Stuart and Bob go to San Francisco to rescue Gru, while Otto follows the trail of a biker who wears an amulet around his neck. When the minions get to the house of Brass Knuckles, they are chased by his henchmen, until Master Cho, a former kung fu teacher and now an employee of an acupuncture clinic, comes to the rescue, fighting off his pursuers. At the request of the minions, Cho decides to teach them kung fu, but they turn out to be incapable students. The trio suddenly and prematurely completes their training and returns to the house of Grandfather Brass Knuckles in order to save Gru. Meanwhile, Otto catches up with a biker in Death Valley, who gives Tom the amulet and gives him a ride to San Francisco...
